2. Schleicher & Schuell BioScience has a library of 60 human and mouse antibody pairs. Our custom array facility will be able to design and build an array for you to meet your needs. 3. Schleicher & Schuell Bioscience's FAST Slides are coated with a 3-D nitrocellulose surface. Nitrocellulose has become the industry standard for protein arraying due to its high capacity and positive influence on protein binding and stability. These surface characteristics provide the foundation for FAST Quant performance as a powerful tool to provide multiplexed data acquisition in a microarray format.
4. The kit contains 4 FAST Slides with 16 pre-arrayed pads on each.
5. FAST Quant is built on standard S&S 16 pad FAST Slides. The slides are standard 25 mm x 76 mm microscope slides so they are compatible with standard microarray scanners. The arrays are 6 mm x 6 mm on 9 mm spacing. Once inserted into the FAST Frame, (offered by Schleicher & Schuell Bioscience and recommended for the use of FAST Quant) the FAST Slides are compatible with the commonly used liquid handling instrumentation as well as with multi-channel pipettors.

S&S highly recommends setting up the dilutions for the standard curve on the first left column of pads in the FAST Frame so as to make it easier to process the slides with ArrayVision FAST. For ease of analysis, incubate the starting concentration of antigens in the uppermost well.

8. The spot density is 36 spots/36 mm2
9. The pitch (center-to-center distance) between two spots is 500 µm.

NOTE: Do not overlap any portion of a pad with the chamber. Do not touch the surface of the pads with anything other than the substances mentioned in this protocol at anytime during the experiment. Take care that pipette tips do not contact the pads and damage them.

  1. The incubation chambers are marked for easy orientation with an indented circle on the upper right corner. The FAST Slides are marked with a laser-etched circle on their upper right corner.

  2. Apply the incubation chamber to the FAST Slide by matching the circle on the FAST Slide with the circle on the chamber. The chamber/slide assembly can then be inserted into the FAST Frame, the end opposite the circle first. Align the assembly with 1 of the 4 positions of the FAST Frame and push it completely into the frame beneath the plastic side rails. Repeat for all chamber/slide assemblies used.
11. As soon as you receive the box containing the antigens and antibodies, open it, retrieve the four vials and place them in a freezer to store at -20C.
12. The detection method used is fluorescence. We recommend the sue of streptavidin-Cy™5 (Amersham Biosciences cat# PA 450001,1 mg/bottle) or an equivalent to Cy5.
